The Endomembrane System
The endomembrane system is a chain of connected organelles that build, modify, package, and ship proteins, with vesicles carrying cargo from one station to the next.
A protein headed out of the cell isn't finished the moment it's built. It needs folding checks, sugar tags, and sorting labels before it's safe to release, so it travels through a production line. The rough ER, studded with ribosomes, is where the protein is made and begins folding. From there it buds off in a vesicle and travels to the Golgi apparatus, which acts like a finishing and shipping department: it adds or trims sugar groups, folds the protein further, and tags it with molecular zip codes. The Golgi then packages the finished protein into a new vesicle addressed to its destination. That vesicle travels to the plasma membrane, fuses with it, and releases the protein outside the cell through exocytosis. Every step happens inside membrane-bound compartments so the protein is never loose in the cytoplasm.
Smooth ER sits next to rough ER on this line but does different work entirely. It isn't just rough ER stripped of ribosomes — it's built with its own enzymes for synthesizing lipids, detoxifying drugs and poisons, and storing calcium ions. Its identity comes from what it does, not from what it lacks.
This system connects directly to the cell's size limits and membrane traffic. Every vesicle that buds off one organelle and fuses with another is moving membrane, not just cargo. When a vesicle fuses with the plasma membrane during exocytosis, its lipid bilayer becomes part of the plasma membrane, so the membrane's surface area increases. Cells balance this by removing membrane through endocytosis elsewhere, keeping surface area roughly steady over time.
Key terms
- Rough ER
- Endoplasmic reticulum studded with ribosomes where secreted and membrane proteins are made and begin folding.
- Smooth ER
- Endoplasmic reticulum lacking ribosomes, specialized for lipid synthesis, detoxification, and calcium storage.
- Golgi apparatus
- Organelle that modifies, sorts, and packages proteins from the ER before shipping them to their final destination.
- Vesicle
- A small membrane-bound sac that transports materials between organelles or to the plasma membrane.
- Exocytosis
- The process by which a vesicle fuses with the plasma membrane and releases its contents outside the cell.
